TKIs do show some good results for treating cancers of less heterogeneity, but only for a while. The fundamental problem with this approach is that tumor colony really has various types of pathways or various cell types to overcome the inhibition of any particular target, even the inhibition itself can be selective and potent.

The ideal paradigm of treating cancer is to really selectively cut off one or more mechnisms that all cancer cells depend, no matter how unstable their genome and how fast they can develop resistance.

Unfortunately with such huge resource devoted to TKIs, funding to other approaches are cutting short. The only way to accelerate research along those lines, in my opinion, might be the way the Australia baby got her cure.
